Medical uses:

Daybue is prescribed for the treatment of Rett syndrome in both adults and pediatric patients aged 2 years and older.

Recommended Dosage and Administration:

  • Dosage: Administer twice daily, in the morning and evening, based on the patient’s weight.
  • Administration: Can be taken with or without food.

Dosage by Patient Weight:

  • 9 kg to less than 12 kg: 5,000 mg (25 mL) twice daily
  • 12 kg to less than 20 kg: 6,000 mg (30 mL) twice daily
  • 20 kg to less than 35 kg: 8,000 mg (40 mL) twice daily
  • 35 kg to less than 50 kg: 10,000 mg (50 mL) twice daily
  • 50 kg or more: 12,000 mg (60 mL) twice daily

Administration Options:

  • Can be taken orally or via a gastrostomy (G) tube.
  • For gastrojejunal (GJ) tube administration, doses must be given through the G-port.

Warnings & Precautions:

Diarrhea: Many patients may experience diarrhea while taking Daybue. Before starting Daybue, patients should stop using laxatives. If diarrhea occurs, they should begin antidiarrheal treatment, increase fluid intake, and inform their healthcare provider. If severe diarrhea or dehydration is suspected, consider interrupting, reducing the dose, or discontinuing Daybue.

Weight Loss: Some patients may experience weight loss. Regularly monitor weight and consider interrupting, reducing the dose, or discontinuing Daybue if significant weight loss is observed.

Adverse Reactions:

The most common adverse reactions (reported in at least 10% of patients treated with Daybue and at least 2% more frequently than in the placebo group) are diarrhea and vomiting.

Drug Interactions:

  • For orally administered CYP3A4-sensitive substrates, closely monitor for adverse reactions when used together.
  • Avoid using OATP1B1 and OATP1B3 substrates concurrently with Daybue if small changes in substrate plasma concentration could lead to serious toxicities.
