CERDELGA is a glucosylceramide synthase inhibitor used for the long-term treatment of adult patients with Gaucher disease type 1 who are identified as CYP2D6 extensive metabolizers (EMs), intermediate metabolizers (IMs), or poor metabolizers (PMs) through an FDA-cleared test.
Limitations of Use:
- -CYP2D6 ultra-rapid metabolizers may not reach sufficient concentrations of CERDELGA for a therapeutic effect.
- -A specific dosage cannot be recommended for CYP2D6 indeterminate metabolizers.
Warning & Precautions
Certain patients should avoid using CERDELGA due to their CYP2D6 metabolizer status, which can increase the risk of side effects, including heart issues. Do not use CERDELGA if you are:
- An Extensive Metabolizer (EM) taking a strong or moderate CYP2D6 inhibitor along with a strong or moderate CYP3A inhibitor, have moderate or severe liver problems, or have mild liver problems and are taking a strong or moderate CYP2D6 inhibitor.
- An Intermediate Metabolizer (IM) taking a strong or moderate CYP2D6 inhibitor along with a strong or moderate CYP3A inhibitor, taking a strong CYP3A inhibitor, or have any degree of liver problems.
- A Poor Metabolizer (PM) taking a strong CYP3A inhibitor or have any degree of liver problems.

CERDELGA can interact with other medications and herbal supplements, potentially increasing the risk of side effects, including changes in heart electrical activity (ECG changes) and irregular heartbeats (arrhythmias).
