Orilissa is a gonadotropin-releasing hormone (GnRH) receptor antagonist prescribed for managing moderate to severe pain due to endometriosis.
Recommended Dosage:
- -For individuals with normal liver function or mild hepatic impairment: 150 mg once daily for up to 24 months, or 200 mg -twice daily for up to 6 months.
- For those with moderate hepatic impairment: 150 mg once daily for up to 6 months.
Administration Instructions:
- -Take Orilissa tablets at the same time each day, with or without food.
- -Use the lowest effective dose, considering the severity of symptoms and treatment goals.
- -If a dose is missed, take it as soon as remembered on the same day, then continue with the regular dosing schedule.
Warning & Precautions
Orilissa can lead to a dose-dependent reduction in bone mineral density (BMD). The longer the medication is used, the greater the BMD loss, which may not fully recover after stopping treatment. Therefore, Orilissa is not recommended for women with known osteoporosis.
Women taking Orilissa at doses of 150 mg or 200 mg may experience changes in menstrual bleeding patterns, including reduced frequency, severity, or duration. This can make it harder to detect pregnancy early. If pregnancy is suspected, perform a pregnancy test and discontinue Orilissa if pregnancy is confirmed.
In clinical studies for endometriosis, some patients treated with Elagolix (Orilissa) experienced suicidal thoughts or behaviors. Patients should seek immediate medical attention if they experience suicidal thoughts or behaviors. The risks and benefits of continuing treatment should be reassessed if such events occur.
Orilissa can cause dose-dependent increases in ALT levels. Patients with elevated liver tests should be promptly evaluated to determine if the benefits of continuing treatment outweigh the risks.
