Before Starting Treatment:
Your healthcare provider will measure your baseline blood phenylalanine levels before beginning treatment with Pegvaliase-pqpz. The initial dose is 2.5 mg, administered as a subcutaneous injection once a week for the first 4 weeks.
Increasing the Dosage:
After the initial 4 weeks, the dosage will be gradually increased over at least 5 weeks. This gradual increase helps your body adjust and aims to reach the target dose of 20 mg once daily. During this period, your healthcare provider will monitor your tolerance to the medication and your blood phenylalanine levels.
Maintenance Dosage:
Once the 20 mg daily dose is achieved, it will be maintained for at least 24 weeks. Your blood phenylalanine levels and dietary intake will continue to be monitored. If, after 24 weeks of 20 mg daily treatment, your blood phenylalanine levels have not decreased by at least 20% or have not fallen below 600 micromol/L, your doctor may increase the dose to a maximum of 40 mg once daily.
Continuing or Stopping Treatment:
If, after 16 weeks on the maximum dose of 40 mg daily, there is still insufficient reduction in blood phenylalanine levels, your healthcare provider may decide to discontinue Pegvaliase-pqpz.
Adjusting Dosage as Needed:
If your blood phenylalanine levels drop too low (below 30 micromol/L), your dosage may be reduced, and your dietary protein and phenylalanine intake may be adjusted to maintain safe levels.
Monitoring and Dietary Adjustments:
